研究概要	無蛋白培養可能なヒト膵がん培養細胞HPC-YOの培養上清からエンドセリン分解酵素(EDE)を発見した。限外濾過濃縮し、凍結乾燥した培養上清(約200L)をゲル濾過、陰イオン交換体、ヒドロキシルアパタイト等のカラムを用いてSDS-PAGE(2ME処理済み)で83Kdaの単一バンドを得た。N末端がブロックされていたため、精製品をリシルエンドペプチダーゼで限定分解し、部分アミノ酸配列を求めた結果、新規酵素であることが判明した。反復測定した結果6ヶ所の8〜12個のアミノ酸配列が明らかとなり、このうち親水性の高いペプチドを2種類合成することが出来た。これらをKLHと結合させ、特異性の高いポリクロナル抗体を作製し、精製EDEを用いて作製したポリクロナル抗体およびモノクロナル抗体を組み合せて、サンドウィッチELISAによる微量測定系を確立しつつある。培養上清を用いた予備実験では30ng/ml〜800ng/mlまで測定出来るようになった。血中濃度測定には末だ感度が低いために螢光プローブを用いた測定系を準備中である。一方、最も重要なcDNAクローニングも2種類の方法で実施している。すなわち、アミノ酸配列から作製したDNA合成プローブを用いたクローニングと、λgt11発現系に特異抗血清を作用させるクローニング法で、ライブラリーにはHPC-YO細胞のmRNAを用いている。
