研究概要

Na,K-ATPaseのαとβアイソフォーム遺伝子のうち全組織でhouse keeping遺伝子として発現するα1、筋組織で発現するα2、神経細胞接着分子AMOGと同一物であるβ2の5′上流制御配列を、変異の導入等によりさらに詳細に解析した。
α1:ARE[-102〜-61]結合タンパク質の検索の結果、ゲルシフト法でC1,C2,C3複合体が観察された。ARE領域をプローブとしたサウスウェスタンクローニングから、別に2種類の結合タンパク質が得られた。その一方であるAREB6は7個のZn-fingerとG1uに富む領域をもち、そのmRNAは心組織や骨格筋に存在するが肝、膵、脾では検出されなかった。他方のタンパク質はすでに他の調節因子として報告のあるHEB(Huet al.,1992)であった。HEBやAREB6のcDNAを培養細胞にトランスフェクトした場合、α1遺伝子の転写への効果は、細胞株や発現量によって正負の両方であった。ARE内にはATF-1結合領域もあり、ATFの結合を確認した。以上のように、ARE領域に6種類以上の蛋白質が結合し複雑に相互作用する全体像が判明した。
α2:Sp1結合配列と類似配列GGGAGGと2個のE-boxを含む領域(-175〜-108)は筋芽細胞L6において数倍の転写促進活性を示した。この領域へのSp1結合による転写促進の様式は、kinetic synergism(Herschlag and Johnson,1993)モデルに適合することを示した。
β2:ラット神経芽細胞腫由来のB103細胞で、特異的制御エレメントAMREをすでに同定していたが、新たに確立したラット脳アストロサイト初代培養細胞や、他の培養細胞株でも、転写制御エレメントとしてのAMREの活性が確認された。AMREに結合するタンパク質のうち一つは、Sp1結合コンセンサス配列を含むオリゴヌクレオチドにより結合が競合阻害される。さらに、抗Sp1抗体による結合阻害がみられた。
