シビレエイ電気器官シナプス前神経終末からのアセチルコリン遊離機構に関する研究

研究概要

(1)シビレエイ電気器官シナプトソームからのアセチルコリン遊離に対する種々のCaチャネルブロッカーの効果を調べた。その結果、アセチルコリン遊離に関与するCaチャネルとしては、N型、P型の寄与が大きいこと、しかしながら、L型の寄与もあることが示された。(2)更に、骨格筋L型Caチャネルのα_2δサブユニットを認識する単クローン抗体(MCC-1)が、アセチルコリン遊離を部分的に阻害することを見出した。(3)MCC-1を固定化した免疫親和性クロマトグラフィーを用いて、MCC-1結合蛋白質を単離精製した。このフラクションをSDS-PAGE及びWestern blot法により分析したところ、非還元条件下に見出される170KDaのバンドがMCC-1結合蛋白質であることが分かった。このバンドは、還元条件下では消失することから、骨格筋L型Caチャネルのα_2δサブユニットと同様の性質を示すことが分かった。(4)MCC-1抗体カラムクロマトグラフィーで精製されたフラクション中に、神経終末細胞膜とシナプス小胞との融合に関与すると考えられているシンタキシンを見出した。(5)並行して、シビレエイ電気器官シナプトソーム膜のCaチャネルの電気生理学的性質を平面再構成膜法により観測する試みを行ったが、未だ成功していない。(6)放射性標識した短鎖リン脂質をプローブとして、リン脂質の外側層から内側層への移行(フリップ・フロップ,translocation)と、平衡分布を測定した。その結果、シナプトソーム形質膜にはホスファチジルセリン特異的なキャリアー(translocase)の存在が示されたが、シナプス小胞膜には、そのようなリン脂質キャリアーは存在しないことが示された。(6)肥満細胞からのヒスタミン遊離(脱顆粒)と細胞内Ca動態の両者を同時測定する手法を開発し、両者の関係を調べることができるようになった。
