胎盤ペプチドホルモン産生細胞の分化に関わるホメオボックス遺伝子のクローニング

研究概要	ホメオボックス蛋白を含め多数の組織特異的転写調節因子が同定され、これらの因子が組織の分化、機能維持に重要であることが示されきた。近年、下垂体における成長ホルモン(GH)、甲状腺刺激ホルモン遺伝子(TSH)の組織特異的発現がPit-1と称される転写調節因子により行われることが明らかにされた。胎盤で合成される胎盤性ラクト-ゲン(placental lactogen)、絨毛性ゴナドトロピン(chorionic gonadotropin)などのペプチドホルモンは、それぞれ下垂体で合成されるGH,TSHと同じ祖先遺伝子から進化したと考えられるため、胎盤でのこれらホルモン産生にもPit-1が関与しているか否かを検討した。RT-PCRを用いた検討により、胎盤では、Pit-1の発現は認められず、他の転写調節因子の関与が示唆された。そこで同じPOUファミリーに属する遺伝子発現を検討したところ、Oct-1の胎盤における発現を確認した。さらにpaired domain遺伝子群に属するPax-8のも胎盤で発現していることを確認し、これら転写調節因子が胎盤での組織特異的遺伝子発現調節に関わっていることが示唆された。一方、Pax-8の活性化に関してはこれまで報告がないためその活性化機構も検討した。その結果、Pax-8は、転写因子JUN、NF-kBと同様、分子内のcystein残基の還元により活性化されることが明にされた。今後、胎盤においてPax-8により調節を受ける遺伝子群を同定したい。
