研究概要

ヒトのサイトカインのレセプターに関してGM-CSFレセプターとG-CSFレセプターを中心として以下の研究を行った.
レセプターに対するモノクローナル抗体の作製:GM-CSFレセプターalpha鎖の遺伝子構造から抗原性が強いと考えられる部分のペプチドを合成し、それに対するモノクローナル抗体を作成した.この高力値のモノクローナル抗体はIgM型で,造血細胞でのGM-CSFレセプター分布をフローサイトメトリーで検出可能とした.フローサイトメトリーで使用可能なG-CSFレセプターを認識する抗体も得ている.
サイトカイン(CSF)の刺激伝達について:G-CSFは幼若造血前駆細胞を増殖させる働きと,成熟した好中球に働きその機能を変化させるという大きく分けて2つの作用がある.各々の作用の刺激伝達には別のkinaseが関与していると仮説を立てて検討した.すると,分裂能を持つ幼若造血細胞にG-CSFを作用させると,成熟した好中球には認められない分子量約130kdの蛋白が新にtyrosine燐酸化されることを見い出した.さらに,この蛋白はtyrosine燐酸化酵素でもあるJAK2であることを同定した.
膜結合型G-CSFレセプターと可溶性G-CSFレセプターの分布および成熟にともなう変化をRT-PCR法で検出した.
さらに,stem cell factorのレセプターであるc-kitやerythropoietinレセプターのフローサイトメトリーによる解析も併せて行った.
