血管内皮細胞におけるトロンボモジュリンの発現調節と血栓傾向に関する研究

研究概要

今回の研究業績の概要は以下の通りである。
1。培養ヒト血管内皮細胞(HUVEC)におけるトロンボモジュリンの発現はトロンビン刺激で一過性(-30分後)にダウンレギュレーションされ、その後4時間目位にピークがくるアップレギュレーションをうける。
2。このHUVECにおけるトロンボモジュリンのトロンビンによる発現制御は新たに発見されたトロンビン受容体を介するシグナル伝達からのものである。
3。トロンビン受容体のアゴニストペプチドを合成して、それでHUVECを刺激しても、トロンビンと同様の効果が観察される。
4。トロンビン受容体を介するトロンビンのシグナルはtyrosine kinaseを使っていることが、インヒビターを使用した実験より判明した。
5。トロンビン受容体の細胞外部分のモノクロナル抗体を作製して、フローサイトメターでトロンビン受容体の測定に成功したが、このトロンボモジュリンと正反対の活性を有する受容体は、内皮細胞のみならず、血管平滑筋細胞、巨核球、血小板膜上に存在した。そしてトロンビン刺激で著明なアップレギュレーションを受けた。これらのことより、トロンビン刺激は内皮細胞などの抗血栓活性を低下させ、血栓活性を増強させることが判明した。
6。冠動脈血栓部位にはトロンビン受容体は著しく強く発現していた。これはこの部位で血栓が生じやすい環境にあることを示唆している。
