GTP結合蛋白質の活性を制御する細胞内因子の検索

研究概要	種々の細胞膜受容体刺激の情報は、αβγサブユニットからなるGTP結合蛋白質(G蛋白質)を介して細胞内へと伝達されるが、それぞれの情報がどのような機構でその情報伝達経路の選択性を発揮しているかについては不明の点が多い。G蛋白質はコレラ毒素や百日咳毒素によりADP-リボシル化されその機能が様々に修飾されるので、情報伝達機構を研究する上で有用なツールとして利用できる。本研究では、この細菌毒素によるG蛋白質のADP-リボシル化反応を利用して以下の知見を得た。 1.G蛋白質のβγサブユニットを固定化したアフィニティーカラムを用いて、種々の細胞の細胞質画分よりβγサブユニットと結合し得る蛋白質を検索した結果、分子量93,000の蛋白質の存在がいくつかの組織、細胞に共通して観察された。2.精製された蛋白質の部分アミノ酸配列や免疫化学的な解析より、この93kDa蛋白質は、ヒートショック蛋白質の一つである90-kDa Heat shock protein(hsp93)と同定された。3.Hsp93は百日咳毒素が触媒するG蛋白質のADPリボシル化を阻害することから、βγサブユニットと結合しその機能を阻害するG蛋白質の新しい制御因子であると考えられた。4.一方、百日咳毒素のADPリボシル化反応を利用して、G蛋白質が細胞膜だけはでなく核内にも存在することが明らかにされた。5.単離細胞核を用いた蛋白質の輸送系を開発し、この核内百日咳毒素基質のG蛋白質が核内へ輸送系に関与する可能性が示唆された。
