研究概要

神経細胞間で信号を搬送する物質を受容する器官の一つに、グルタメートリセプターがあり、これは記憶や学習など重要な高次機能に関係していると考えられている。本課題に於いては、この受容体の機能について有機合成の側から解明研究を行った。
1)本受容体に作用するカイノイドはC_4側鎖にπ電子を持つことが強い活性を示す一つの条件であることを見出しているが、このπ電子をもつ系はピロリジン環と直交したコンホメーションで働いていると考えられる。そこで逆に側鎖のπ電子系がピロリジン環と同一平面上にあるような化合物(1〜4)を合成して活性を調べた。合成はトリメチレンメタンが分子内のαβ-不飽和エステルに付加することにより、3-アザビシクロ[3.3.0]オクタン骨格を作る方法で行った。二重結合の位置は金属触媒によって調整した。脱分極活性は全てについて著しく減り、やはり側鎖のπ電子平面がピロリジン環と直交するコンホメーションが強い活性を示すために必要であることを示唆した。
2)C_4に芳香環をもつカイノイドの新しい短段階合成法を開発した。4-ヒドロキシプロリンから4-フェニル-3-デヒドロプロリンを合成し、これにマロン酸モノメチルエステルのマンガン(III)酸化で生ずるラジカルを付加させ、酢酸基がC_4に生じたOII基とラクトンとなっている形のフェニルカイノイドを立体選択的に合成した。これを水素添加してフェニルカイノイドとした。また、この方法によってC_4にピリジンをもつカイノイドの合成にも成功した。
3)3-デヒドログルタミン酸にトリメチレンメタンを付加させてグルタミン酸にexo-メチレンシクロペンタンを作りつけた化合物CPG-を合成し活性を調べた。4個の立体異性体中CPG-IVのみがカイニン酸同様の活性を示した。
