変異ミトコンドアDNAを導入したトランスジェニックマウスの作製法の開発

研究概要	変異mtDNAを導入するにはmtDNA分子を細胞に導入するのが最善であるが再現性に問題があることが判明したので、再度細胞質とmtDNAを失った細胞の融合によってmtDNAを導入することにした。今年度用いたのは、心筋症患者由来のミトコンドリアtRNA-Ileに点変異をもつ細胞とAlpers病由来のミトコンドリアtRNA-Argに塩基置換をもつ細胞である。これらの細胞より核を除去しmtDNAが完全にない細胞と融合させた。そして、これらのサイブリドの性質を調べた。心筋症由来の患者の変異mtDNAを持つサイブリドではミトコンドリア内でのタンパク質合成が低下したのに比べ、Alpers病由来のmtDNAを持つ細胞では正常細胞とくらべ変化はなかった。呼吸鎖酵素活性も正常で心筋症由来の細胞では呼吸鎖酵素活性は低下していた。また、ミトコンドリアの形態もAlpers病由来mtDNAを持つサイブリドは正常であった。このことより、Alpers病由来のmtDNAの塩基置換は病的なものではなく、希な多型によるものであることが判明した。この技術によって、病的な塩基置換と個人差である多型の区別ができるようになった。また、老化とmtDNAの関係についても研究が進められた。老人より分離した線維芽細胞では呼吸鎖活性が低下していたが、mtDNAが完全にないHeLa細胞に老人由来のmtDNAを移入させるとタンパク質合成能も呼吸鎖酵素活性も完全に回復した。そのため、老化による呼吸鎖酵素活性の低下はmtDNAの変異によるものではなく、核遺伝子に原因があると推定できた。
