研究概要

単一細胞由来の遺伝子ライブラリーの作製法を確立し、固有の遺伝子の単離もしくは、その単一細胞での遺伝子発現パターンの解析を行うことを最終目的とした。
(1)去年までは細胞ではなく、それから抽出した核酸であるRNAを出発材料として、いかに微量のRNAからスタート出来るかという点を中心に技術の開発を進めて来て0.5pg程度のRNAからcDNA全体を増幅できることが判明していた。今年度は出発材料をRNAではなく細胞から直接cDNA合成を行う技術開発に焦点を絞って行った。
始めは、0.5%NP-40を用いて細胞をLysisバッファーに入れた後、65℃で1分、室温で3分処理後すぐにcDNA合成を行ったが、細胞のLysisが不完全なため、cDNA合成が出来なかった。その後、SDSやTriton-X100などを使用したが、結果は同じであった。そこで、細胞は完全に溶解していなくても部分的に細胞膜に穴が出来ているはずなので、cDNA合成や、PCRに必要な試薬を細胞内に染み込ませる時間を設けてIn-Cell PCR法の様に反応を行ったところcDNA合成/増幅に成功した。つまり全体として細胞からRNA抽出という過程を経ずに直接cDNA合成/増幅が可能になった。その感度を調べたところついに1個の細胞から直接cDNA合成/増幅が出来ることが判明した。つまり当初の本研究の目的を達したことになる。
(2)現在はこの方法を用い、極微量にしか存在しないVα14^+T細胞のリンホカイン産生パターンなどの検討を行っている。
(3)またこの方法の変法として、cDNA合成の際に遺伝子特異的プライマーを用いることにより、目的の遺伝子由来の転写産物のみを1個の細胞から増幅して解析する方法を現在開発中である。
(4)この方法の応用のひとつとして、細胞の分化段階特異的なcDNAを単離するためにサブトラクションcDNAライブラリーを作製する方法も現在開発中である。ビオチン化プライマーとStreptavidineを使う方法を用いることにより比較的簡単にサブトラクションcDNAライブラリーを作製できることがわかった。
また去年まではcDNA全体の増幅法として、inversePCRにこだわっていたが、この環状化法を用いなくてもAnchorPCR法を用いれば、もっと簡単にcDNA全体の増幅可能であることが分かり、その方法に切り換えた。
