研究概要	ヒトロタウイルスの複数の血清型に対し防御能を有し、腸管免疫を効果的に賦与し、そして副作用が少ないヒトロタウイルスワクチンを目指して、ポリオウイルス人工DIベクターにヒトロタウイルスVP4遺伝子の交叉反応性中和エピトープを含む抗原部位をコードする領域を挿入し、そのキメラウイルスの免疫原性を検討してきた。この過程で、いくつかの課題が生じた。交叉反応性中和エピトープを含むVP5′の抗原性から意外と低く、むしろPタイプ特異的なエピトープに富むVP8′が強い抗原性を示したことから、VP8′+VP5′を発現することとした。また、免疫沈降反応およびウエスタンブロッティング法で、発現蛋白の性状を抗VP4抗体、抗ビリオン抗体を用いて検討したが、明確には同定とすることができなかった。そこで、あらためてバキュロウイルス発現VP4(およびVP8′とVP5′)に対するより高力価の抗血清を準備するとともに、ベクターの改良を進めている。さらに、ポリオウイルスレセプター遺伝子を導入したトランスジェックマウスについては、経口感染が十分に効率的でないのでこの点についても改良を進めている。一方、VP4の中和エピトープをさらに詳細に調査すべく、各種抗VP4中和モノクローン抗体とユニークな性状を有するヒトロタウイルスおよび各種動物ロタウイルスとの反応性について検討し、ヒトロタウイルスVP4がヒト特異性を有することを明確にした。また、多数のヒトおよび動物ロタウイルス由来VP4遺伝子を解析し、ヒトと動物間のVP4遺伝子の組換えの可能性も示した。
