試験管内「血液脳関門」モデルの作製と新しい脳特異的輸送担体開発の試み

研究概要

1.星状グリア細胞が血管内皮細胞を脳型に「トランス分化」させることが見い出された。すなわち、初代培養ラット胎児脳星状グリア細胞のフィーダー層でヒト臍静脈内皮細胞を培養すると、γ-glutamyl transpeptidase、transferrin receptor、P糖タンパク質といった脳血管内皮特異形質が誘導され、[^3H]イヌリンに対する「血液脳関門」脳も獲得された。
2.これらの脳血管内皮特異形質の発現は、星状グリア細胞が産生する細胞外マトリックスタンパク質と分泌性タンパク質を介し、遺伝子の転写レベルで誘導されることが明らかにされた。
3.ヒト肺動脈内皮細胞と平滑筋細胞でI型ミネラルコルチコイドレセプターをコードするmRNAが検出され、また、alternative splicingの結果生じたと推定される新しいミネラルコルチコイドレセプターmRNA分子種も発見された。さらに、アンジオテンシンIIによる血管平滑筋細胞の肥厚が、アルドステロンで増強され、I型ミネラルコルチコイドレセプターの特異的拮抗剤であるZK91587で抑制されることが見い出された。これらの知見から山本らは、「血管アルドステロン」が実際血管局所で機能し、血管壁の再構築や高血圧症の発症に関係している可能性を提唱した。
4.山本らはまた、血管内皮増殖因子(vascular endothelial growth factor、VEGF)mRNAに相補的なアンチセンスオリゴヌクレオチドを血管内皮細胞に移入し[^3H]チミジンの取込みを調べた結果、低酸素下での内皮細胞のDNA合成が著しく抑制されることを見い出し、「血管VEGF」のオートクリン/パラクリン作用という血管新生の新機序を提唱した。
