研究概要	GM-CSF主に骨髄細胞の増殖と分化を促進する。血球細胞のGM-CSFレセプターの発現と分化との関係を検討するため、血球細胞分化のすべての段階でヒトGM-CSFレセプターを構成的に発現するトランスジェニックマウスを作製した。GM-CSFの活性は種特異的であるので、これらのトランスジェニックマウス由来骨髄細胞中の造血前駆細胞のヒトGM-CSFに応答する増殖分化をメチルセルロースコロニー形成法によって解析した。マウスGM-CSFは顆粒球マクロファージ(GM-)コロニーを形成するのに対し、ヒトGM-CSFはGM、好酸球、肥満細胞、赤芽球、巨核球、blast細胞、ミックスコロニー等の様々なタイプのコロニーの形成を支持した。従って、コロニー形成に対するヒトGM-CSFの効果はマウスGM-CSFよりはマウスIL-3のそれにより近かった。さらに、ヒトGM-CSFはblast細胞コロニーやミックス細胞コロニーをマウスIL-3よりも多く生成した。ヒトGM-CSFはさらにエリスロポエチン非存在化でも赤芽球コロニーを形成した。従って、ヒトGM-CSFはヒトGM-CSFレセプターを発現してさえいれば、前駆細胞からすべての系統の造血細胞を誘導する能力を持っていることが明らかとなった。
