バチルス属細菌の高分子型・低等電点型の新規セリンプロテアーゼの研究

研究概要	本研究はネパールの伝統的発酵食品キネマより分離したビオチン要求性細菌が、從来既報のBacillus属の生産するセリンプロテアーゼとは分子量、等電点の大幅に異る新規のセリンプロテアーゼを生産することを発見したことにはじまる。 1.生産菌の分離同定: ネパールのキネマより分離したプロテアーゼ生産菌No.16を同定したところ、対照株である納豆菌Bacillus subtilis(natto)と同様の性質を示すことからBacillus subtiles(natto)と同定された。 2.酵素の分子特性: 分離菌を培養すると分子量約28,000、等電点8.7のスブチリシンNATと同様なセリンプロテアーゼの外に、分子量はHPLCで88,000、SDS-PAGEで約90,000、等電点は3.9のセリンプロテアーゼを生産した。從来の知見からはbacillopepかdase Fは分子量50,000あるいは33,000で多少近い。精製酵素の比活性は0.034katal/kg・タンパク質でミルクカゼインに対する最適pHは10.0である。本酵素の内部構造の25残基のアミノ酸配列はbacillopeptidase FのN末端と相同性があった。 3.基質特異性: 過ギ酸酸化インスリンB鎖に対する特異性は、pH7.5でTyr^<16>-Leu^<17>を最も良く分解し、さらにCgSO_3H^7-Gly^8、Val^<12>-Glu^<13>、Phe^<25>-Tyr^<26>の各ペプチド結合を加水分解するものであった。この酵素の特異性は從来から知られたスブチリシン・ファミリーのセリンプロテアーゼの特異性とは全く異るものであることを明かにした。本酵素はスブチリシン・ファミリーを特異的に阻害する低分子の阻害剤PMSF、キモスタチンには阻害されたが、タンパク性阻害剤であるStreptonyces subtilisin inhibitorによっては全く阻害されない特性を示した。
