渦鞭毛藻及び緑藻を用いたジメチルプロピオテチンの生合成に関する遺伝子工学的研究

研究概要	揮発性有機硫黄化合物である硫化ジメチル(DMS)は、海洋における独特の匂い、いわゆる「磯の香」の香気主成分であり、のりなどの緑藻類や渦鞭毛藻類、あるいはオキアミ、エビあるいはカワハギなどの海産物中によく見いだされる。また、陸上植物のトマトや茶などにもこの物質の存在が報告されている。最近、このDMSが地球規模の硫黄循環に重要な役割を果たしていることが明かとなり、特に海洋からの大気中への硫黄のフラックスにはこの物質が関与している。海洋におけるDMSの分布はクロロフィルa量とよい相関を示し、一次生産力の高い海域ではDMS量も多いことも明らかにされている。DMSは、陸上の高等植物ではS-メチルメチオニンスルフォニウムが前駆体であることが報告されているが、藻類におけるDMSの前駆体はジメチルプロピオテチン(DMPT)であり、DMSはDMPTの酵素的分解あるいはアルカリ処理によってアクリル酸とともに生成される。本研究では、このDMPTの生体内での役割を明らかにすることを目的として、DMPTの生合成経路を明らかにし、さらに遺伝子工学的にこの物質を有効利用とするものである。 DMPTを大量に生産する渦鞭毛藻Crypthecodinium cohniiを材料として用いた。DMPTはシステイン、メチオニンを経て合成されること、メチオニンからDMPTへの経路は脱炭酸、脱アミノおよびメチル基付加の3反応からなることを明らかにした。続いて、DMPT生合成の第1段階を触媒するメチオニン脱炭酸酵素を完全精製し、その諸性質を調べ、さらにN-末端アミノ酸配列を決定した。本酵素の分子量は約230KDaで、約100KDaのサブユニット2個から構成されるホモダイマーであった。本酵素の性質を検討したところ、Km値は16.3mMであり、至適pHは7.3、至適温度は50Cであった。本酵素の活性におよぼす金属イオンの影響を調べたところ、Zn^<+2>で阻害され、Mg^<+2>で僅かに活性化された。補酵素としてはピリドキサール燐酸を要求した。本酵素のN-末端アミノ酸をエドマン法により決定したとこ、Ala-Leu-X-Trp-Ser-Asp-Ile_Ser-Arg---であった。
