延髄孤束核による脳循環調節の脳内神経経路とその神経伝達物質

研究概要

平成5年度は、延髄孤束核(nucleus iractus solitarius,NTS)よりの脳血管調節の脳内神経経路を明らかにするため、尾側腹外側降圧野(caudal ventrolateral depressor area,CVL)と吻側腹外側昇圧野(rostral ventrolateral pressor area,RVL)との脳循環調節の脳内神経経路を調べた。
1.CVLをL-glutamate(100nl,1.7nmole)の微量注入により化学刺激し脳血流の変化を調べた(n=9)。刺激と同側の大脳皮質の血流量は64±9より48±9ml/100g/min(mean±SE)に有意(P<0.01)に低下し、脳血管抵抗は1.7±0.2より2.4±0.4mmHg/〔ml/100g/min〕へと有意に(P<0.01)上昇した。
2.両側頸部交感神経を切断した後1の実験と同様の方法でCVLを化学刺激し脳血流の変化を調べると、脳血流量・脳血管抵抗は有意な変化を示さなかった(n=10)。
3.muscimol(500pmole)を一側RVL(交感神経の脳内origin)に注入しその部の神経細胞のみの活動を阻害した後、1の実験と同様の方法でCVLを化学刺激し脳血流の変化を調べた(n=11)。脳血流量・脳血管抵抗は有意な変化を示さなかった。
以上の結果よりCVLによる脳循環の調節はRVLと頸部交感神経を介している事が判明した。RVLもCVLと同様に脳血管収縮機能を持っている事が私達により既に明らかにされているので、CVLよりRVIへは脳循環に関しては交感神経興奮性経路がある可能性が示唆される。また、NTSよりCVLへ交感神経興奮性経路がある事は既に多くの研究者より明らかにされているので、NTSによる脳血管調節はその脳内神経経路としてCVLとRVLを介し、頸部交感神経を通じて行なわれている事が示唆された。
