GATA転写因子群による赤血球系細胞特異的な遺伝子発現調節機序の解析

研究概要

本研究の目的は、赤芽球分化の際にGATA転写因子群が一群の遺伝子の発現を活性化する機序を解明することであり、さらにそれを通して、各GATA因子が赤芽球分化に果たしている役割を明らかにすることにある。目的を達成するために、本研究では 1)各GATA因子をコードする遺伝子のクローン化と、その調節領域の構造と機能の解析、及び 2)種々の遺伝子調節領域に存在するGATA配列に実際に結合しているGATA因子の分子種の同定を行った。前者に関して、マウスGATA-1、ヒトGATA-2およびヒトとニワトリGATA-3遺伝子の構造を詳細に決定したところ、これらGATA群をコードする遺伝子の構成は各因子間で、また、種を越えてたいへんよく保存されていた。また、ニワトリGATA-3遺伝子のプロモーター上流域にはGATA配列がCACCC配列とクラスターを作って存在し、この部位はTリンパ球では抑制的に、一方、赤芽球では促進的に機能していることを見出した。後者の課題に関して、マウス赤白血病細胞核をフォルムアルデヒドで固定後、GATA-1/DNA復合体を抗GATA-1抗体を用いて免疫沈降し、得られた画分からDNAを抽出した。この中を調べてみると、赤血球特異的に発現しているGATA-1自身の遺伝子やエリスロポエチン受容体遺伝子に存在するGATA配列は存在するものの、赤血球には発現していないT細胞受容体遺伝子のGATA配列は見出されなかった。すなわち、同じGATA配列の中でもGATA-1因子の結合する配列は細胞によって限定されており、このことから、本因子の結合がこれらの遺伝子発現に重要な貢献をしていることが示唆される。これらの研究を通して、GATA転写因子群が細胞・組織特異的な転写調節に機能していること、また、その発現が巧妙に調節されている様子がさらに明らかになった。
