潰瘍性大腸炎一大腸癌モデルに関する実験病理学的研究

研究概要

1-HydroxyanthraquinoneをACIラットに1%の濃度にて飼料投与し、投与開始後4、8、12ヶ月にて屠殺し、盲腸、上部結腸、下部結腸粘膜上皮の細胞増殖性を粘膜上皮の厚さ、BrdUの標識率、ornithine decarboxylase(ODC)活性の測定によりassayし、又潰瘍性病変、陰高膿瘍、メラノーシス等の変化を経時的に検索した。 陰高長は経過と共に延長し、その傾向は盲腸、上部結腸に於いて強かった。BrdUの標識率では陰高長と類似せる傾向を示し、1-hydroxyanthraquinoneの投与を受けたラットでは対照群に比し、4、8、12ヶ月のいずれの時点でも高い標識率を示した。とくに12ヶ月時ではその傾向が顕著であった。ODC活性は盲腸、上部結腸、下部結腸のいずれに於いても4、8、12ヶ月時共1-hydroxyanthraquinoneの投与ラットが対照ラットよりも有意に高かった。メラノーシスの出現は盲腸では4ヶ月よりみられ、結腸では8ヶ月より認められた。陰高膿瘍の出現は盲腸では8ヶ月よりみられ、結腸では12ヶ月より認められた。潰瘍の形成は盲腸では8ヶ月よりみられ、結腸では12ヶ月より認められ、一般にその程度は盲腸に於いて強かった。以上の1-hydroxyanthraquinone投与ラットに於ける大腸粘膜上皮の経時的観察により、腫瘍発生部位に一致して粘膜上皮細胞の増殖性の亢進があることが分り、細胞増殖性のhydroxyanthraquinone発癌に於ける重要な意義が明らかにされた。又、潰瘍性病変の出現部位と腫瘍発生部位もほぼ一致し、同病変の出現と発癌との密接な関係も示唆された。
