潰瘍性大腸炎-大腸癌モデルに関する実験病理学的研究

研究概要

1)1,8-Dihydroxyanthraquinone(danthron)は長期間の経口投与により,びらん,炎症細胞浸潤,粘膜上皮の再生肥大などの潰瘍性大腸炎類似の毒性変化を来たし,その後,大腸腫瘍を誘発する。Danthronのマウスにおける大腸毒性および発癌性に関して,他の大腸発癌物質の発癌性に対する促進性を中心に解析した。6週齢の4群のICR/CD-1マウスを使用し,第1群(30匹)には実験開始後の12週間に,週1回1.2-dimethyLhydrazine(DMH)を20mg/Kg皮下注射した。動物は基礎食CE-2にて維持された。第2群(30匹)はDMHの投与後42週間にわたってdanthronを0.2%の濃度にて飼料投与された。第3群(30匹)は生食を12週間,週1回投与された後,danthronを飼料投与された。第4群(30匹)は無処置対照群とされた。第2群における結腸腫瘍(腺腫,腺癌)の発生率および多発性はいずれも第1群に比し,有意に高かった(70%:41%,p<0.05)2.52±2.72:0.69±0.99,p<0.01)。盲腸における腺腫様過形成はdanthronの投与を受けた第2,第3群にのみ発生し,その発生率はおのおの100%であった。第1,第2群では肛門部の扁平上皮癌がおのおの14%,22%発生した。Danthronの投与を受けたマウスの結腸粘膜上皮ではオルニチン脱炭酸酵素の活性が有意に上昇した。潰瘍性大腸炎類似の組織学的変化はdanthronの投与を受けたマウスの盲腸及び結腸に限ってみられた。以上の様な成績はdanthronがマウスにおいてDMH誘発大腸発癌をみずからの標的臓器である結腸において促進し,肛門部位では相互作用を示さないことを示す。
2)ハムスターにおいて1-hydroxyanthraquinoneの長期毒性試験を1%飼料濃度にて2年間投与の実験にて行った。1-Hydroxyanthraquinoneはハムスターにおいてはラットと異なり,明瞭な腸管の慢性毒性や発癌性を示さなかった。このことはhydroxyanthraquinoneの腸管毒性に対する種属差を示すものである。
