細胞傷害性T細胞による細胞内寄生体の排除に関する分子機構

研究概要	C57BL/6およびBALB/cマウスを用いてBCGに対する細胞傷害性T(Tc)細胞を誘導し、BCG感染マクロファージを標的細胞として6時間および18時間細胞傷害試験を行なったが、IL-2でTcを増幅させても十分なキラー活性を認めることはできなかった。つまり、Tc細胞はBCG感染に対する防御機構としては大きな役割を担っていないと考えられた。最近、我々はBCGの排除にはマクロファージによって産生される一酸化窒素(NO)がその重要な働きをすることを確認した。NOはIFN-γなどの刺激によって誘導的に発現するNO合成酵素(NOS)によって合成されるため、NOSの発現が細胞内寄生体の排除に大きく関与している可能性がある。NOSmRNAの発現はLPSやTh1細胞などにより産生されるIFN-γによって誘導され、Th2細胞によって産生されるIL-4やIL-10によって抑制される。そこで、これらの抵抗性(B10)及び感受性マウス(BALB/c、C3H)のNOSmRNA発現の制御を理解するためにBCGに対するサイトカインmRNAの発現様式を検討した。その結果、1)B10ではNOS,IL-2,-4,-12,IFN-γのmRNAが検出された。これに対し、BALB/cではNOS,IL-12,IFN-γのmRNA発現が低下し、C3Hではこれらに加えさらにIL-2及びIL-4のmRNAは検出されなかった。C3HではTh1,Th2共にBCGに対する反応性が低いことが示唆された。2)マクロファージのNOSmRNAの発現能を組換えIFN-γ刺激で調べたところ、B10ではこれら全てに強い発現が認められたが、BALB/c及びC3Hでは弱かった。3)IFN-γはマクロファージにIL-12mRNAの発現を誘導することが判明したが、B10に比べBALB/c及びC3Hでは低下していた。以上により、BALB/c及びC3HではIFN-γ産生能の低下に加え、マクロファージのIFN-γによるNOS産生能の低下が加わり、BCG感受性を示すと考えられる。マクロファージによって産生されるIL-12はTh1細胞を誘導することが知られているが、我々はIFN-γがIL-12mRNA発現を誘導すること、BALB/c,C3Hではこの誘導能が低いことを見出した。このことはIFN-γ→マクロファージ→IL-12→Th1→IFN-γというPositive regulatory circuitが感受性マウスでは弱いため、抵抗性マウスに比べ、最終的に顕著に低いNOSmRNA発現を示すと考えられる。
