LPS刺激によりマクロファージ内で誘導されるplastirリン酸化経路の解析

研究概要	(1)LPS刺激持異的にリン酸化される65‐kDaの基質蛋白質(PP65)の部分アミノ酸配列は、既に決定されている。この配列を基にオリゴDNAを合成し、マクロファージ(Mφ)mRNA由来のcDNAを鋳型として用いたPCR法によりPP65をコードする遺伝子断片を増幅し、そのDNA配列(約300bp)を決定した。この配列はヒトのl‐プラスチンとアミノ酸レベルで約95%の高い相同性があり、マウスにおけるプラスチンであることが確定した。プラスチンは、Ca^<2+>結合ドメイン、カルモジュリン結合ドメイン、アクチン結合ドメインをもつユニークな蛋白質で、ヒト線維芽細胞の癌化に伴って出現する蛋白質として注目され、最近同定された新種の蛋白質である。ヒトT細胞をIL‐2で刺激した時にリン酸化されることも、最近明らかにされた。LPS刺激でこの蛋白質が特異的にリン酸化されることは、LPSのシグナル伝達を明らかにする上で重要な手がかりになると考えられ、興味深い。(2)次に、PP65をリン酸化するプロティンキナーゼ(PKE:Protein Kinases induced by endotoxin)について検討した。LPS刺激Mφの蛋白質をSDS‐PAGEで分画し、PVDF膜に転写後、7Mグアニジンで復元し、膜上でリン酸化反応を行った(renaturation kinase assag)。この方法で、82‐55‐,45‐KDaの分子量をもつ少なくとも3種類のPKEがLPS刺激によって活性化されることが明らかになった。複数のリン酸化酵素の関与を証明した報告はこれまでなかった。このうちのどれがPP65をリン酸化するのかはまだ明らかではないが、既知の代表的キナーゼとは異なるようである。LPS刺激によって複数のPKEが活性化されること、そしてそれからが多くの基質をリン酸化することは、LPSの多彩な生物活性を説明する根拠となるかもしれない。そのうちのPP65は一次構造が決定され、ユニークな機能をもつことが明らかにされた。
