研究概要

1.ラット気管支上皮のクララ細胞が分泌する新しいセリン型酵素トリプターゼ・クララを分離同定し、これが肺における野生株ウイルスの活性化プロテアーゼの本体であることを証明した。更に、トリプターゼ・クララが気道上皮にのみ分布することがセンダイウイルスの感染増殖が肺に限局することの直接の原因であることを証明した。
初感染巣である気管支上皮細胞において、ウイルスの出芽極性が尖頂領域に限局していることが気道における局所感染を規定し、一方、側基底領域から出芽することが全身臓器へのウイルスの播種に必要であることを証明した。
センダイウイルスに感染した上皮性細胞が膜融合を起こして融合巨細胞を形成するためには、F蛋白の開裂活性化に加えて、ウイルス糖蛋白が側基底領域に発現していることが必要条件である。
側基底領域からも出芽するF1-R株の感染細胞では、微小管などの細胞骨格系の構造と機能が破壊されており、上皮性細胞の持つ細胞極性が破綻していた。更に遺伝子塩基配列の比較から、F1-R株の変異M蛋白がこれに関与していることが強く示唆された。
