肝炎ウイルス遺伝子発現の細胞分化誘導シグナル伝達系と癌抑制遺伝子転写への影響

研究概要	発癌プロモーターであるテレオシジンは肝癌細胞の増殖を抑制し、形態を著明に変化させる。このテレオシジン作用のシグナル伝達には通常のプロテインキナーゼCとは構造的に異なると考えられるプロテインキナーゼCが関与することを見い出した。このプロテインキナーゼは、おそらく細胞内の小胞体のカルシウムイオンチャンネルのリン酸化を介してカルシウムの細胞質への動員を抑制している可能性が示唆された。このことは肝癌細胞の分化誘導にプロテインキナーゼCやカルシウムが密接に関与していることを示唆している。またテレオシジンはc-fosやp53の発現を促進することも示された。従って癌遺伝子と考えられるc-fosの発現の促進が直接癌化の促進に関与するとは考えにくい。同じ発癌プロモーターであるタプシガルジンは、c-fosやp-53の発現を促進するなどテレオシジン作用と類似した作用をもつ。細胞増殖を抑制するとともにテレオシジン作用に括抗し、細胞骨格蛋白の重合や空胞状構造の形成を阻害する。タプシガルジンは小胞体カルシウムATPaseの特異的阻害剤であり、小胞体による細胞質カルシウムの調節が細胞の形態や機能に重要な働きをしていることが示された。一方、肝炎、肝硬変、肝癌患者からC型肝炎ウイルスRNAをRT-PCR法で検出し、ウイルスの非構造領域や構造領域の増巾を行なってきた。ウイルスのエンベロープ領域やN2、N3領域を増巾し、また肝癌からDNAを抽出し、PCRでα-スエトプロティンエンハンサーを増巾した。これらの遺伝子断片を発現ベクターPRC/CMUやレトロウイルスベクターに組み込む作業を行っている。
