ラット小腸におけるコレシストキニン遺伝子発現の制御とその放出の機序

研究概要

CCKにかぎらず、生理活性ペプチドは生体内で重要な役割を担っているにもかかわらず、その合成と分泌に関する制御機構はあきらかでない部分が多い。覚醒ラットでは腸管内から膵液と胆汁をのぞくと、小腸壁からCCKが血中の分泌されて膵外分泌が亢進するという特徴がある(luminal feedback regulation)。我々はその特徴を用いてCCK分泌や合成の制御機構を検討してきた。その結果、これまでに2種類のCCK放出因子の存在があきらかになっている。小腸由来のCCK放出因子はまだその本体があきらかになっていないが、膵液由来のCCK放出因子はmonitor peptideとしてすでにアミノ酸構造もあきらかにされ、膵臓の腺房細胞で合成されることが証明された。Monitor peptideはラットの持つ2種の膵分泌性トリプシンインヒビター(PSTI)-56と-61のうち-61とおなじものである。そこで膵液と胆汁を上部小腸から遠位回腸へバイパスし、小腸内CCKmRNAの変動を検討するとともに、PSTI-56,61のmRNAを定量し比較した。結果はCCKmRNAはバイパス1日目より増加し、28日目まで持続した。そして小腸内CCK濃度は漸増し、28日目で最大となった。しかし血中CCK濃度は7日目で最大となり、14日から28日目にかけて減少した。PSTIのmRNAも同様に増加したが、興味深いことに(通常ではPSTI-56mRNAのほうが多いのであるが)、7日目まではPSTI-61mRNAの増加が著しく、PSTI-56mRNAよりも高値を呈した。しかし14日目以降はPSTI-56mRNAのほうが高値となった。本年度は膵管結紮による膵実質萎縮状態での十二指腸内CCKmRNAの変動を検討するとともに、PSTI-61の膵臓内での遺伝子発現とその制御機構を明らかにし、CCKの合成、分泌の制御機構との相互関係をあきらかにしたいとかんがえている。
