ラット小腸におけるコレテストキニン遺伝子発見とその放出の機序

研究概要

1。CCKmRNAの定量法の確立
RT-PCR法により増幅して作製したCCKcDNAfragmcntをブローブとして、ラット小腸、脳、および心臓にCCKmRNAの存在を確認することができた。slot-blot analysisとbeta-actinmRN Aとの比率により、小腸CCKmRNAの増減の算出が可能となった。
2。血中CCK濃度とCCKmRNAとの関係
覚醒ラットを用い、〓液胆汁を小腸内から除去した場合、血中CCK濃度は1-2時間目に最大となり、4時間目以降は軽度に減少した。CCKmRNAは、4時間目に有意の増加がみられ、24時間目にはふたたび減少した。セクレチンmRANレベルもCCKmRNAの変化とほぼ平行して変動がみられた。
3。CCK-releasing peptideのひとつであるPSTI-61の〓臓内mRNAレベルとCCKとの関係
〓液胆汁を上部小腸から下部小腸にバイパスすると、1日目には血中CCK値が著名に上昇し、3、7日目まで上昇が持続した。PSTI-61mRNAレベルは、1回目に上昇し、3日目に最大となったが、小腸内CCKmRNAは1日目から上昇はするが、7日目に最大となった。
4。〓管結〓時の血中CCK、組織内CCKmRNA、PSTI-61mRNAの相互関係。
〓管を結〓してしまうと、血中CCKは上昇するが〓実質は萎縮する。血中CCK値は,28日間にわたって、軽度上昇をしめし、大きな変動はみよれないが、小腸内CCKは、7日目に最低となり、14日目にovershootし、28日目に前値に復する。CCKmRNAレベルは、組織濃度に先行して変動した。しかし、セクレチンmRNA値は全く変動しなかった。PSTI-mRNAは、3日目に最大となり、以後減少した。
以上のことから、CCKの合成〓進(mRNAレベルの増加)は、CCK分泌〓進に伴っておこること、またCCK分泌〓進刺激はすなわち合成〓進刺激になりうると結論される。〓由来CCK放出因子(PSTI-61)の遺伝子発見は、血中CCKにより制御され、またCCKそのものの遺伝子発見も部分的にCCKにより制御されていると推測される。
