冠血流調節における内皮由来弛緩因子およびエンドセリンの役割に関する研究:in vivoモデルによる検討

研究概要	冠血流量の調節における内皮由来一酸化窒素(EDNO)の役割を麻酔犬を用いたin vivoの実験系にて検討し、L-アルギニン類似物質であるモノメチル-Lアルギニン(LNMMA)がEDNO生成阻害剤として有用であること(J Cardiovasc Pharmacol 1991;18:665-669)、EDNOが一過性冠閉塞に続く反応性充血に関与していること(Am J Physiol 1992;263(Heart Circ Physiol 32):H8-H14)を既に報告した。さらにはより強力なEDNO生成阻害剤であるニトロ-L-アルギニン(LNAME)を用い、冠動脈抵抗血管においてそのトーヌスの調節にEDNOの基礎分泌が大きく関与している(Arch Int Pharmacody 1994;in press)が、一方、血管平滑筋が受動的収縮刺激を受けた場合はEDNOが反応性に放出されることはないことも明らかにした(J Cardiovasc Pharmacol 1994;in press)。さらに本研究ではEDNOとスーパーオキシドとのin vivoでの相互作用を明らかにする目的で、8-フェニルテオフィリンにてアデノシンの作用をブロックした麻酔犬において、スーパーオキシドディスムターゼ投与前後の反応性充血を比較検討し、一過性虚血〜再灌流中にはスーパーオキシドが生じ、EDNOを不活化することにより反応性充血を減弱させていること、また反応性充血におけるEDNOの役割は主にその後半部分に認められることを明らかにした(第1回アジア微小循環学会)。これらの事実より、EDNOは冠血流量の調節に重要な役割を果たしており、さらにその作用はスーパーオキシドとの相互作用により影響されることが示された。
