チロシナーゼ陰性型白皮症の病因であるチロシナーゼ遺伝子の異常についての研究

研究概要	先天性白皮症のうち、チロシナーゼ(以下Taseと略す)陰性型においては、染色体TaseDNAの塩基配列の変異により色素細胞内に活性のない不完全なTaseが作られ、それが病因となることを我々は世界に先駆けて発表した。今までに我々が日本人の患者3人において見い出した異常は、Tase遺伝子の蛋白coding regionの一塩基挿入によるnonsence mutation(Biochem Biophys Res Commun 164:990,1989)とpoint mutation(J Biol Chem 265:17792,1991)である。 Tase遺伝子のプロモーター領域の異常により、RNA転写が起こらず、そのためTaseの蛋白合成がなされないために発症する白皮症もあるはずである。しかし、このような症例の報告はまだない。このような症例では、今までの様なTase蛋白のcoding regionの異常の検索をいくら行っても決して、その病因を明らかにできない。そこで今回の研究では、プロモター領域の異常の検索も行えるようなシステムの開発を試みた。そのためにまず、正常なプロモター領域の解析を行った。その結果、ヒトTaseのプロモター領域として、転写開始点より上流までの約200塩基が得られた。しかし、これはマウスのプロモター活性と比べて3-4倍低い。そのため、ヒトのTase遺伝子においては、発現を促進するようなElementが、もっと上流領域に存在する可能性が示されました。事実、Tase遺伝子の転写開始点よりも約1.8kbから2.0kbの間約200bpに、SV40プロモターのコントロール下で色素細胞特異的なエンハンサー活性を検出した。さらに、この約200bpを解析した結果、エンハンサー活性に不可欠であった39bPのコアエレメントが同定された。今後、Tase陰性型患者のうち、Tase蛋白のcoding regionの異常の見い出せない場合は、Tase遺伝子のプロモターおよびエンハンサー領域の異常の検索を行う予定である。
