薬物モデルを用いた精神分裂病症状の発現に関する脳内神経回路の研究

研究概要

初年度に引き続いて、分裂病様症状惹起薬を投与したラットの脳における最初期遺伝子c‐fosの発現パターンとその発達に伴う変化を調べた。一部の実験では、ドーパミン(DA)代謝の変化もあわせて検討した。成熟期では、metham phetamine(MAP)とcocaineの急性投与によってc‐Fos様免疫反応陽性細胞が脳の広い範囲にわたって出現するが、そのパターンは、DA受容体アゴニストであるapomorphineと類似していた。Phencyclidine(PCP)はこれらとは異なり、NMDA受容体アンタゴニストdizocilpineと同様のパターンを示すことがわかった。また、MAP投与後のc‐Fosの発現は、DA受容体アンタゴニストの前処置によって抑制された。MAP、cocaineおよびPCPによる脳内c‐fos遺伝子の発現パターンは、生後発達に伴って変化し、特に大脳新皮質における変化がもっとも著しかった。生後8日の幼若ラットにおける発現パターンを比較すると、MAPとcocaineはapomorphineに類似し、PCPはdizocilpineに類似していた。一方、成熟期のラットにおけるPCP投与後の細胞外液中DA量の増加は、前頭葉で著しく線条体では軽度であり、神経インパルスフローに依存性であったのに対し、MAPによる増加は双方の脳部位で著しく、インパルスフローには依存しなかった。
以上の結果から、c‐fos遺伝子の発現を指標として、MAPおよびcocaineは、PCPとは異なる脳内神経活動の変化を引き起こすことがわかり、これらの薬物による臨床症状の違いを反映すると考えられた。また、c‐fos遺伝子発現の生後発達の研究は、薬物による分裂病様症状の発現が、大脳新皮質の神経回路の発達と関係する可能性を示唆している。さらに、DA遊離の実験でも、MAPとPCPとの間に作用差が認められ、c‐fos発現の差異とも関係すると考えられる。MAPおよびPCPの投与ラットに共通して認められる前頭葉のDA伝達亢進は、抗精神病薬(DAアンタゴニスト)で改善される症状関係する可能性がある。
本年度はこのほかに、抗PCPおよび抗MAP作用を示すD‐セリンについての研究行った。D‐セリンは、ラットばかりでなくヒトやマウスにおいても脳選択的に分布する内在性物質であり、その濃度は大脳皮質で高く小脳では痕跡程度であること、ヒト大脳皮質において胎生期から老年期まで高濃度に保持されることなどを明らかした。
