男性不妊の新しいマウスモデルを用いての、不妊症の免疫調節療法の基礎と開発

研究概要

男性不妊症の新しいモデルとして、同系精巣細胞(TC)のみの皮下注射により、C3H/Heマウスに実験的自己免疫性精巣炎(EAO)を発症させる系を確立した。この新EAOモデルを免疫制御の検定系として、免疫制御に係わる2種類のサプレッサーT細胞株を樹立した。1つはEAOの前臨床期抑制性CD4^+T細胞株(C.Ts)で、細胞性・液性両免疫反応と病変を抑制し(Clin.Exp.Immunol.1992)、もう1つはEAOの誘導期に作用して発症と細胞性免疫反応を抑制するCD8^+T細胞株(Ts‐A)である(Clin.Immunol.Immunopathol.1992)。本年度の成果としては、
1.抑制性T細胞とT細胞レセプター(TCR)α鎖の関係が谷口ら(1989)、Dorfら(1991)により示唆されているので、前臨床期抑制性CD4^+T細胞株(C.Ts)のTCRα鎖のusageをPCR法、M13シークエンス法により解析した結果、productive,nonoroductive共にVα5であったが、Jαは両者間で異なっていた(谷口らとの共同研究)。
2.EAOの誘導期抑制CD8^+T細胞株(Ts‐A)はTCRαβを有しており、現在、TCRα鎖の解析を行っている。
3.EAO惹起性エフェクター細胞株を樹立した。BALB/cマウスにTCを4日おきに2回腹腔内注射後採取したリンパ節、脾細胞を、マウス部分精製精子抗原(mTA)とcyclosporineと共に培養し、さらに、X線照射feeder細胞の存在下で、mTAによる選択を行った。樹立された細胞株(B.T.1)は1‐A^+のCD4^+T細胞で、正常BALB/cマウスに5x10^5個を腹腔内注射すると全例に精巣上体炎を伴うEAOが発症した。B.T.1細胞の特異抗原(mTA)刺激培養上清中には、マクロファージ遊走阻止活性が認められた(Int.J.Androl.1992)。本細胞株は特異抗原であるTCに対しキラー細胞活性を示さなかったが、正常マウス皮膚から分離した線維芽細胞を抗原非特異的に傷害した(Andrologia,1993)。
