DNAトポインメラーゼを標的とする婦人科癌化学療法の基礎的および臨床的研究

研究概要

DNA topoisomerase-I(Topo-I)は一本鎖DNA切断を触媒するとされている。Topo-Iと切断複合体を形成する抗癌剤はカンプトテシンとその誘導体しか知られておらず、基礎的、臨床的研究も未だ充分ではない。今回、培養細胞においてRT-PCR法を用いてTopo-I mRNA活性を半定量的に解析し、新規のカンプトテシンであるCPT-11に対する細胞毒性と比較検討した。又、微小管に作用する新規の抗癌剤paclitaxelに関しては培養細胞中にてギャップ・ジャンクション(GJ)の構成蛋白であるコネキシン(Cx)43の発現誘導を観察した。
[材料と方法](1)Caov-3,HHUA,Ishikawa,HeLa S3の4種の培養細胞からRNAを抽出し、RT-PCR法により、cDNAの標的部位を増幅し、エチジウムブロマイド染色したPCR産物をデンシトメトリカルに解析し、その活性を求めた。そして、そのTopo-I mRNA活性をCPT-11に対する細胞毒性と比較した。また、通常のSouthern blot法により、その結果の正当性を確認した。
(2)Ishikawa株にpaclitaxelを投与し、GJの構成蛋白であるCx43の発現誘導を観察した。
[結果](1)上記4細胞系はCPT-11に対して感受性を示し、その細胞毒性はTopo-I mRNA活性レベルと相関した。その結果はSouthern blot法で確認された。
(2)培養細胞系であるIshikawa株においてpaclitaxelにより、本来認められなかったCx43の発現誘導が認められた。
[結語]RT-PCR法を用いたTopo-I mRNA活性の測定方法は微小検体から検出可能であり、個々の症例に対してTopo-I活性の有無を検討し得て、臨床応用可能であると考えられた。
GJは未分化な癌細胞や細胞回転の活発な場合に少なくなることから、paclitaxelは培養細胞系に分化誘導を導き、細胞間communicationを促進するものと考えられた。
