C-Kitおよびそのリガンドによる卵成熟・子宮内膜・胎盤の分化調節機構の解析

研究概要

1.マウス卵・初期胚におけるc-kitの発現:c-kit蛋白に対するモノクローナル抗体を用いた免疫組織化学染色により以下の知見を得た。(1)卵巣においては顆粒膜細胞や莢膜細胞においてc-kitの発現は認めず、卵の膜表面にのみその発現を認めた。(2)卵における発現をさらに詳細に検討すると、胎仔期卵巣では認めず、出生後の卵巣においてのみその発現は認めた。また、排卵刺激による発現の推移をみると、排卵後に急激に発現が減弱した。(3)初期胚における検討では、受精直後の1細胞期に最も発現が強く、細胞数の増加とともに減弱し、4細胞期以後には消失した。
2.マウス子宮内膜・胎盤におけるc-kitの発現:(1)c-kit蛋白に対するモノクローナル抗体を用いたwestern blottingにより、妊娠子宮および胎盤にc-kit蛋白の発現を認めた。(2)免疫組織化学染色によりc-kitの発現の局在を検討すると、子宮内膜においては妊娠以後に初めたc-kitの発現を認め、妊娠初期においては子宮内膜腺管上皮表面に限局していた。ただし、luminal epithrliumには発現を認めなかった。妊娠中期以降になると腺管上皮における発現は徐々に減弱し、uterine crypt周囲の間質や胎仔から離れた場所の脱落膜に発現が認められた。妊娠後期の成熟胎盤においてはlabyrinthine layerおよびdecidual layerに強い発現を認めるが、giant trophoblastic layerおよびspongiotrophoblastic layerには認めなかった。また、labyrinthine layerにおいては、母体血に接するtrophoblast cell表面にその発現が限局していた。(3)northern blotting,reverse transciptase polymerase chain reactionによりc-kit mRNAの発現を検討したが、上記結果と整合する結果が得られた。
以上の知見から、c-kitは卵・初期胚の成熟およびそれらをとりまく環境である子宮内膜・胎盤の分化と密接に関連した蛋白であることが推察された。
