新規分子ツール群の開発と応用による心筋イオンチャンネルの分子機能の解析ー不整脈病因の解明をめざして

研究概要	1.L型Ca^<2+>チャンネルに対する代表的なブロッカーである1.4-ジヒドロピリジン化合物の結合部位を初めて同定した昨年度の骨格筋標品での成果をかまえ、本年度は心筋標品を用いてその検索を進めた。その結果、ビヒドロピリジン化合物の結合部位は心筋標品においても骨格筋同様、ドメインIIIとドメインIVの2ヶ所の特定部位であることを明らかにした。またこの2ヶ所のアミノ酸配列を心筋と骨格筋で比較したところ、数個所で、アミノ酸置換がみられることから、この置換が両組織におけるビヒドロピリジン化合物に対する親和性の差を決めていると推定された。 2.心筋Na^+チャンネルをターゲットとする抗不整脈薬メキシレチンの結合部位を光アフィニティラベル法によって明らかにするため、そのラベル用試薬の調整を行った。またラベル部位同定のためのツールとして抗ペプチド抗体2種と抗リガンド抗体を作製した。 3.ATP感受性K^+チャンネルの分子的同定をめざし、トリチウム標識したグリベンケラミド型光ラベル試薬を合成した。この試薬は心筋のATP感受性K^+チャンネル標品とナノモーラーオーダーの高い親和性を有することが確認できた。この試薬を用いて光ラベル反応を行ったところ、K^+チャンネルタンパク候補となる分子を特異的にラベルすることができたので、これを用いて現在このタンパク分子の同定を進めている。
