悪性リンパ腫でのIL-6とその関連遺伝子発現の解析と新しい治療法の確立

研究概要	non-Hodgkin's Iymphoma(NHL)のFollicular large 4例, Follicularmixed 1例, Diffuse large 8例, mixsd 3例, medium 3例, small 1例につき、 IL-6, IL-6R, 及びgp130の発現をreverse transcriptase-PRC法を用いて、測定した。ポジティブコントロールとして、 IL-6, IL-6R, 及びgp130を発現するヒト・ミエローマ細胞株U266の発現量を1.0として、各症例の発現量を比較した。 IL-6の発現は、 Folliculor lymphoma 5例中3例、 Diffuse large全例、mixed 1例、 medium 2例、 small 1例に見られた。特にDiffuse large(B cell type)の1例では、 U266に比し、 2.3倍の発現が見られた。 IL-6Rは、 Diffuse mediumの1例以外の全例で発現していたが、全般的に発現量は少なかった。gp130は、全例に発現しており、U266と同程度の発現を示すものから、50分の1程度のものまで、種々であった。 IL-6, IL6R, 及びgp130の発現量に関し、それぞれ2つの間には、相関は見られなかった.U266の2.3倍のIL-6を発現していたDiffuse largeの症例では、 IL-6R及びgp130の発現はともに低くいため、 autocrine growthは考えにくいと思われる。以上のことより、 NHLでは、 IL-6, IL-6R及び gp130の発現は、全般的に低く、 NHLの発現、進展に対するIL-6の関与を積極的に示唆する結果は得られかった。
