造血因子の情報伝達系を介する白血病細胞の増殖機構

研究概要	骨髄性白血病細胞の自律性増殖機構を解明するためにはまず造血幹細胞の増殖がどの様に制御されているかを明らかにすることが重要である。我々は正常血球や急性骨髄性白血病より樹立されたヒトGM-CSFやIL-3依存性の細胞株(M07E)を用いた検索によりGM-CSFやIL-3受容体を介する情報伝達系には共通の細胞質内蛋白(P93やP70など)のチロシンリン酸化や癌原遺伝子産物Raf-1のセリンリン酸化と活性化ばかりでなくMAPキナーゼ(p42,p44)のチロシンリン酸化と活性化が関与していることを明らかにした。現在、これらのシグナル伝達分子が白血病細胞の異常増殖にどの様に関与しているかを検討中である。また、M07E細胞などの白血病細胞株や多くの急性骨髄性白血病(AML)細胞はチロシンキナーゼ型受容体の一種である癌原遺伝子c-kit遺伝子を発現しており、一部の白血病細胞では、c-kitリガンド(stem cell factor:SCF)により増殖ならびに分化が促進されることを明かにした。c-kitはリガンド(SCF)との結合により活性化され、自己リン酸化、基質〔フォスファチジルイノシトール3キナーゼ(P13キナーゼ)など〕との結合などの諸反応が惹起され血球にシグナルが伝達される。各種ヒト白血病細胞や細胞株を用いてc-kitの活性化の有無について検討すると、約半数のAML症例では、AML細胞はSCF刺激のない状態でも軽微ではあるがc-kitのチロシンリン酸化、活性化が認められた。また、ヒトマスト細胞白血病細胞では、SCFを全く産生していないにもかかわらず、SCF刺激前より強度なc-kitの恒常的チロシンリン酸化と活性化が認められ、c-kit遺伝子に点突然変異が認められた。現在までに報告されているc-kitの遺伝子異常はすべてc-kitのチロシンキナーゼ活性を低下させるものばかりであったが、我々の知見は、c-kitは正常造血ばかりでなくヒト白血病細胞の増殖や分化、あるいは腫瘍化にも関与している可能性を示唆するものである。
