研究概要

tRNAのアミノアシル-tRNA合成酵素によるアミノ酸受容特異性の研究対象として、プロリンtRNAを選び、in vitroの系で検討した。その結果、大腸菌のプロリンtRNAのプロリル-tRNA合成酵素による認識部位として、アンチコドン2文字目と3文字目のG35G36、アンチコドンに隣接するG37、識別位塩基A73、TPSIステム中のG49、アクセプターステムのC1-G72(特にG72)およびG3-C70塩基対、それにジヒドロアーム中のU17Aを同定した。プロリル化活性に影響を与えたこれらの部位が真にプロリル-tRNA合成酵素によるポジティブな認識部位であることを確認するために、大腸菌のトレオニンtRNAにこれらの部位を順次導入する事により、ほぼ完全にプロリンを受容するように特異性のスイッチングが起こり、先に同定した部位の塩基および塩基対が真にプロリル-tRNA合成酵素による認識部位である事を立証した。また、大腸菌では唯一RNAレベルで一次構造が知られていない3種のプロリン tRNAの内、メジャーなプロリンtRNA(アンチコドン、CGG)をDNAプローブを用いて単離し、修飾を含めて一次構造の決定をおこなった。
