研究概要	(1)ニワトリ・リゾチームの老若マウス肝細胞における分解半減期老若マウス肝細胞に導入したニワトリ・リゾチームの分解半減期はそれぞれ約60時間、と約30時間であった。すなわち、老齢では半減期が2倍に延長していた。 (2)酸化修飾リゾチームの老若マウス肝細胞における分解半減期ニワトリ・リゾチームを鉄イオン触媒によって酸化し、老若マウス肝細胞に導入して分解半減期を調べたところ若齢で約16時間、老齢で約20時間だった。すなわち、いずれの細胞でも酸化リゾチームの方が酸化していないリゾチームより速く分解された。分解は、老齢で若干遅くなっているが老若の間に有意差は見られなかった。 (3)酸化蛋白質を優先的に分解する酵素の精製ラット肝臓から酸化リゾチームを非酸化リゾチームよりよく分解する酵素を精製した。精製酵素の性質を詳細に検討したところ、これは従来からインスリン分解酵素IDEとして知られているものに一致した。IDEが酸化蛋白質を優先的に分解するというのは新たな発見である。ラット肝臓ホモジネートのIDE活性(蛋白質あたり)は、酸化リゾチームを基質にした場合老若でほとんど変わらず、インスリンを基質にした場合は老齢でやや(20〜30%)低下していた。 (4)老若ラットの大脳皮質および脳幹におけるプロテアソーム活性異常蛋白質の分解に関わっているとされているプロテアソームの活性測定法をラット脳抽出液を用いて確立し、老若ラットの大脳皮質および脳幹のプロテアソーム活性を測定したところ大脳皮質では老齢の方が30%低かったが脳幹では逆に30%ほど高かった。プロテアソームの活性で見る限り大脳皮質では加齢で異常蛋白質が蓄積しやすくなるといえる。
