研究概要

主要組織適合性抗原遺伝子(MHC)は、胸腺内でT細胞レパートリーの決定に関与し脊椎動物免疫系における自己と非自己の識別を分子レベルで確立する。また個体が寄生体の感染を受けたときには、外来抗原の提示をT細胞にし免疫系の初期反応を律する働きをする。高畑は、チュービンゲンでの共同研究を通じ、MHCの機能を集団遺伝学の立場から定量的に研究した。この結果は、MHCの多型的な遺伝子座の数が種によらず6〜7個と一定に近いことを説明し、T細胞および結合ペプチドの両レパートリーの大きさとMHC多様性の程度を結びつけたものである。MHCの異常な多様性は、そのT細胞の胸腺内での拘束と関係していることが定量的に明らかになった。MHC分子の高次構造や提示ペプチド分子に関する知見、またMHC分子の人種内における多様性や病気との関連に関する知見の集積には目ざましいものがある。こうした最新の知見をまとめる作業を颯田と共同で行ない、LABFAXに発表した。この本はMHC研究を実験的に進める上で有効な参考書である。マックス・プランクのヤン・クラインとは、2冊の本を執筆している。1つは、彼が編集している本の一章で、MHCに関する集団遺伝学の理論と実験データを紹介するものである。他の1つは、人類の進化に関する本で、来年ケンブリッジ大学から出版予定である。マ-シ-・ウエノヤマとは、植物における自己・非自己識別分子の共同研究、マサトシ・ネイとは、集団遺伝学の理論的研究を進めた。この2人には、昨年夏にあった国際植物会議(横浜)で高畑がオ-ガナイズしたシンポジウムに招待講演を依頼し、引き続き一週間程共同研究を行なった。分子進化の基本的原理は、中立説によって説明できることが多いが、動植物を問わず自己・非自己識別分子の理解には適用できないことが明らかになりつつある。
