研究概要	本年度はおもにマクロファージアシアロ糖タンパク質レセプター(M-ASGP-BP)の糖鎖認識の解析を重点的におこなった。本レクチンの糖鎖結合特異性をY.C.Lee博士(Johns Hopkins University)より恵与された合成糖鎖ペプチド(cluster glycosides)をリガンドとして用い検討した。反応系は生理的に近い条件として、本レクチンcDNAをCOS-1細胞にトランスフェクトして細胞表面に発現させ、このM-ASGP-BPとアシアロオロソムコイドとの結合に対する各種合成糖鎖ペプチドの阻害活性を測定した。その結果、M-ASGP-BPは従来より良く研究されている肝臓のアシアロ糖タンパク質レセプター(H-ASGP-BP)とはかなり異なった特徴的な糖鎖結合性をもつことが明らかとなった。具体的には、H-ASGP-BPは実はガラクトースよりもN-アセチルガラクトサミンに高い親和性を示すのに対し、M-ASGP-BPはN-アセチルガラクトサミンよりもガラクトースに対して高い親和性を示す。H-ASGP-BPはいわゆるゴールデントライアングルと呼ばれる位置に3個のガラクトース残基が配置したトリアンテナ型の糖鎖に非常に高い親和性を持つのに対してH-ASGP-BPはこのトリアンテナ型よりもより大きな空間に配置した3個のガラクトース残基に対して一層高い親和性を示した。この結果は、H-ASGP-BPが主として血液内の糖タンパク質に含まれる秩序立った構造を持つ糖鎖をリガンドとするのに対して、M-ASGP-BPはガン細胞表面の糖鎖を含めた生体異物としての脱秩序型の多様な糖鎖との結合が可能であることを示している。マクロファージの持つ生体異物排除機能の一つとしてM-ASGP-BPの役割は興味深い。
