肝細胞増殖因子(HGF)とc-metレセプターによるがん化機構に関する共同研究

研究概要	1、種々のがん細胞におけるHGF及びc-met遺伝子の発現異常の解析種々のヒト上皮系がん細胞でHGFの産生を検索し、ヒト肺がん細胞SBC-5細胞がHGF産生能を持つことを見出した。このHGF分子種は、第2クリングルドメインまでを含むHK_1K_2型変異体であることを明らかにした。この細胞はc-metレセプターも発現していることから、オートクリンループの形成により細胞運動性が亢進され、細胞の浸潤、転移性の獲得といった癌細胞の悪性化に関与していることが示唆された。 2、HGF人工変異体の作製とレセプターへの結合及び活性化能の検定。種々の人工変異体を作製し、c-metレセプター及びヘパリンへの結合能と活性化能を測定した。その結果、HGFのα鎖内のヘア-ピンループ構造及び第1、第2クリングルドメインが、これらの活性発現に必須であることを明らかにした。N末端から第2クリングルドメインまでを含むHK_1K_2型変異体はc-metに結合するが増殖活性はなく、HGFのアンタゴニストとして作用する可能性を示した。 3、HGF変換酵素の精製とその阻害剤の開発 HGFは不活性型前駆体プロHGFとして細胞外に分泌され、その後、変換酵素により活性型2本鎮HGFとなる。本研究ではHGF変換酵素を精製し、さらにこの酵素を特異的に阻害するペプチドクロロメチルケトン誘導体を合成し、その阻害作用は明らかにした。この阻害剤はHGFの活性化を阻害するので、ある種の癌細胞の増殖を阻害する可能性が考えられる。
