研究概要

HLAトランスジェニックマウスを用いたin vitroのシステムを用いたHLAの機能解析、HLA分子に結合するペプチドの検索、及びHLA遺伝子の構造決定を行い、HLAによる免疫応答、疾患感受性の制御について以下の知見が得られた。
1)HLAクラスII分子として、HLADQ6のみを発現しているHLA-DQ6/I-A Knockoutマウスを作製したところ、成熟CD4T細胞の分化が認められたことより、マウスT細胞かDQ6分子により正の選択を受けることが直接的に証明された。
2)6種のHLAクラスI分子に結合しているペプチドをHLA分子より単離し、構造分析を行い、それぞれのHLA分子に特有なモチーフ明らかにした。HLA分子上に1〜2アミノ酸の変化により結合ペプチドのモチーフが大きく変化することが証明され、これがHLAによる免疫応答の差異の分子構造であると考えられた。今回明らかになったモチーフは、HLA-A*0201結合ペプチドが2残基目Lcu、C末Val、Lcu,A*0206が2残基目Val、Gln、C末Val、Luc,A*0207が2残基目Lcu,3残基目Asp、C末Luc,A*0204は2残基目Lcu,C末Lcu,HLA-B51が1残基目Asp、2残基目Ala、Pro、C末llc,HLA-B52が2残基目Gln、C末llcであった。
3)HLA-A遺伝子座のDNAタイピングのシステムがセットアップされ、いくつかの新しいHLA-A遺伝子が同定された。また、HLA-A*0206とA*0207は異なる疾患感受性を示すことが明らかになり、両者が異なるペプチドと結合することがその分子機構であると考えられた。
