研究概要	1. ヒト胎盤より精製したチミジンホスホリラーゼが血管新生活性を有するか否か、チミジンホスホリラーゼの酵素活性が血管新生に必須か否かを調べた。実験系としては受精鶏卵の漿尿膜(CAM法)、マウスの皮下にゼラチンスポンジを移植したもの(ゼラチンスポンジ法)を用いた。精製チミジンホスホリラーゼは、CAM法、ゼラチンスポンジ法のいずれにおいても高い血管新生活性を有することが明らかとなった。ゼラチンスポンジ法でチミジンホスホリラーゼの血管新生を測定する時に、チミジンホスホリラーゼの特異的阻害剤である6-アミノ-5-クロロウラシルをマウスに経口投与したところ、チミジンホスホリラーゼの血管新生はほぼ完全に阻害された。このことは、この血管新生がチミジンホスホリラーゼによるものであり、酵素活性が血管新生に必須であることを示している。ボイデンチャンバーを用い、チミジンホスホリラーゼが血管内皮細胞に対して遊走能の亢進を引き起こすか否かを調べたところ、遊走能の亢進が見られた。 2. チミジンホスホリラーゼの酵素活性に必須な部位に相同性のあるPD-ECGFcDNAの部位の一塩基置換突然変異体2個をPCR法により作製し、COS発現ベクターに挿入した。各々をCOS細胞にトランスフェクトし、チミジンホスホリラーゼ活性を調べたところ、一塩基置換突然変異体のいずれにもチミジンホスホリラーゼ活性は認められなかった。これらのPD-ECGFと一塩基置換突然変異PD-ECGFの血管新生活性をマウスゼラチンスポンジ法で調べたところ、PD-ECGFでのみ血管新生活が認められた。これらの実験結果より、血管新生にはチミジンホスホリラーゼ酵素活性が必須であることが明らかになった。
