脈管作動性ペプチドの分子認識機構と生理作用の研究

研究概要

平成5年度は脈管作動性ペプチドの構造解析、生理作用、受容体、および生体内分布の研究を進めた。PACAPおよびPRPを化学し、その構造を解析した。CDスペクトルおよびNMRスペクトルによる構造解析の結果、PACAP38の9-26位と28-38位にはαヘリックス構造が存在し、20-21位にはヒンジが存在することが明らかとなった。PACAP27の構造は、C端部のヘリックス構造を欠く以外、PACAP38と同じであった。両分子型ともN端部は自由な構造をとっていた。この構造解析に基づき、各種誘導体を化学合成し、その生物活性を検討した。PACAP38および27は、麻酔犬の大腿動脈およびモルモットの単離血管標本に於いて用量依存性の強力な血管拡張作用を示した。PACAPの血管拡張作用は内皮細胞およびNOに依存せず、直接作用であり、PACAP38の作用は27より持続性があった。この持続作用にはPACAP38のC端の構造が関係していた。即ち、PACAP37、36、35、34、33と順次C端を短くすると作用時間は減少したが、最大反応量には変化が無かった。PACAPのN端のHis残基はこのペプチドの作用発現に重要であった。PACAPの生体分布と生合成を研究するために、PACAPおよびPRP特異的ラジオイッムノアッセイ系を確立し、モルモットに於いて両ペプチドの組織含量ならびに分布を調べ、このペプチドが視床下部のみならず消化管など各臓器に広く分布していた。受容体の構造と分布を検索するために、セクレチンならびにエンドセリン受容体のcDNA構造を基にその部分構造を化学合成し、家兎に免疫することにより膵管細胞ならびに肝の伊東細胞と特異的結合する抗体を得、本法が有用であることが示された。
