研究概要

DNAウイルスが感染後、細胞DNAプログラムを誘導するまでの過程を発がんの初期過程として、また細胞増殖誘導のモデルとして、さらには、細胞遺伝子発現調節のモデルとして転写調節に的を絞って解析した。
1.ウイルス初期遺伝子の発現:
細胞の分化の状態は、ウイルス初期遺伝子の発現に大きく影響し、ウイルス感染の許容状態あるいはウイルス発がんに関与する潜伏感染の成立に係わっている。アデノウイルスの系では、E1A遺伝子による細胞遺伝子発現調節が発がん機構の焦点となっているが、E1Aがウイルス遺伝子を活性化する際に働く転写因子が精製され、そのcDNAが単離された(半田)。またE1Aに働く転写因子も明らかにされた(吉田)。パピローマウイルス、B型肝炎ウイルス、EBウイルスは組織特異性が強く、その特異性を決定する因子の解明を進めた。安本はパピローマウイルスの系で、上皮細胞特異的転写因子、高田(等)は、肝炎ウイルスXプロモーターに働く因子、高田(賢)は、EBウイルスの潜伏感染に係るプロモーターBZLF1のプロモーター構造を明らかにした。
2.ウイルス初期遺伝子転写調節機能の細胞側標的:
アデノウイルスE1Aは、MHCクラスI抗原の発現を抑制し、癌遺伝子c-junを活性化する。それらの遺伝子に働く転写因子が明らかにされ(横山、石井)、SV40の系では、T抗原により発現誘導される細胞表面抗原の遺伝子のプロモーター領域がクローニングされた(山口(宣))。パピローマウイルスにおいては、初期遺伝子の発がん機能の解析が進められ(吉池、清野)、山口(政)は増殖細胞核抗原遺伝子を正・負に調節する因子およびその遺伝子の解析を進めた。
