研究概要

本研究の目的は、個々の器官、腫瘍の放射線リスポンスの特異性に基づき、よりよい時間的線量配分(可能なれば有効薬剤の併用も含め)を理論的に導き出すための基礎研究を行なうことにあり、1.腫瘍・正常組織の生物学的特異性に基づく時間的線量配分(分割照射)の決定に重要な種々の因子の解析、2・腫瘍治癒と正常組織障害の先行指標の新たな開発、を長期的な目標とする。この1年間は、特に、腫瘍細胞の増殖能・放射線感受性の測定法を中心に先行指標に関する基礎研究に比重を置いた。1)Micronucleusによる放射線感受性の定量。小野らは、分野照射で重要な低線量域(3Gy以下)での統計誤差を減らしたコロニー形成率測定法を確立し、Micronucleus Assayと比較検討し、Micronucleus Assayの問題点についても明らかにした。安藤らは、再酸素化がPLD修復と放射線感受性に及ぼす影響を検討し、PLD修復が感受性に重要という立場を裏づけた。2)宿主側の制癌効果を含めた免疫組織化学的方法(小川)。これには、腫瘍細胞の悪性度や増殖能に関連すると考えられている癌関連遺伝子,Ki-67,PCNAなどの単クローン抗体(増田、安藤、小川、鈴木)を含め、放射線治療効果の予測指標としての有用性を検討してきた。癌関連遺伝子や単クローン抗体による増殖能や細胞動態の解析は方法が確立され、症例を集めている段階であり、一方、放射線応答におけるその意義について培養細胞を用いた基礎的検討も進んだ。自然経過や手術療法において一般的にいわれる悪性度と放射線感受性や放射線療法の予後とは全く別ものである。一方、3)細胞死・アポトーシス(新部、安藤、鈴木)及び4)小線量照射による正常組織の放射線抵抗性誘導(佐々木)に関しても研究が進んだ。鈴木らは照射によって誘導される新しいタンパク質(p41)、佐々木はリンパ球染色体異常誘発に関する抵抗性の誘導について解析を進めた。
