研究概要	c-kitレセプターチロシンキナーゼは種々の細胞に発現しているが、メラノサイト、マスト細胞、赤血球、生殖細胞の分化・増殖には、c-kitレセプターチロシンキナーゼを介したシグナル伝達が必須である。本班では主に血液細胞と生殖細胞に目標をしぼり、単独あるいは班員同士の協力のもとに研究を行なった。(1)北村はc-kit遺伝子座の多数の突然変異の分子生物学的異常と表現型の間の相関性についてしらべた。表現型としては皮膚のマスト細胞と毛色を対象とした。c-kit突然変異の性質と表現型の間には、はっきりした相関性がみられた。日本で発見されたc-kit突然変異W^nについては、北村と西宗が協力して、W^n遺伝子座がコードしているc-kit蛋白質の性質をしらべた。W^nはチロシンキナーゼ領域の点突然変異であるが、W^n/W^nマウスより由来する培養マスト細胞ではc-kitが細胞外に発現されず、c-kitチロシンキナーゼの点突然変異の多くがdominant negativeの表現型をとるのに、W^nはnull表現型を示した。(2)金倉は北村と協力して、ヒト、マウス、ラットのマスト細胞腫ではc-kitレセプターチロシンキナーゼが、リガンドであるstem cell factorと結合しなくても活性化していることを発見した。これらのマスト細胞腫株ではチロシンキナーゼ領域に共通の点突然変異を持ち、c-kitを発現していない細胞へのトランスフェクションによりこの点突然変異が、c-kitの活性化の原因であることが明らかになった。(3)西川はc-kitの第1イントロンにエンハンサー部位を同定した。(4)松居は生殖細胞で発現しているいくつかのキナーゼのcDNAをクローニングした。これらのキナーゼにはいままで報告されていないものも含まれていたので、その性格づけがなされつつある。
