細胞増殖因子からみた癌細胞と間質細胞との相互作用 -その分子機構-

研究概要

本年度成立した本研究班は焦点をかなり絞った結果、1年目で多くの新しい研究成果が報告された。即ち、c-metの発現を示す癌細胞からの増殖因子・サイトカインと間質線維芽細胞からのHGFとのパラクリンループが存在し、胃癌の形態形成・浸潤に重要な役割を担っていることが示された。また、HGF産生促進因子として、1L-1α,TNFαに加えて、プロスタグランシン-E1,-E2およびグリコサミノグリカンが見いだされた。一方、HGFの第3クリングル以降を欠くHK1K2バリアントHGFがオートクリン的に肺小細胞癌細胞株SBC-5の接着・浸潤を促進していることが明らかにされた。その上、上記バリアントと異なるHGFトランスクリプトが多くの消化器癌に広く発現していることとが認められ、HGF産生は間質細胞のみならず癌細胞で起きている可能性が示唆された。一方、ラット初代培養肝細胞が産生分泌するリン酸型fetuinがHGFと結合して、HGFの肝細胞増殖作用を抑制することが見いだされ、リン酸型fetuinはいわゆる肝のchaloneである可能性が考えられた。
更に、EGF受容体の細胞外領域とc-metの細胞内領域からなるキメラ受容体およびその変異体(ATP結合部 K1108Aおよび主要自己リン酸化の変異体 Y1233F)を作成し、マウス・メラノーマ細胞1G-F1にトランスフェクションして、キメラ受容体およびその変異体発現細胞のEGFに対する反応性を解析した結果、c-metの主要自己リン酸化部位のチロシンリン酸化が細胞分散、運動性亢進作用のシグナル伝達に必須であることが示された。
一方、エズリン、ラディキシン、モエシンからなるERMファミリーが、CD44に直接結合するという大変興味ある所見が見いだされた。
