研究概要	1)細胞増殖におけるプロテアーゼ機能の解析:Rasがん細胞から大量に分泌されているカテプシンLの前駆体に増殖促進作用があることが確認された。プラスミノゲンアクチベーターの発現と癌の悪性度、転移形式に密接な関連がみられた。筋細胞で転写因子のレベルの調節に関与する新しいカルパイン分子種の構造と機能相関を解析した。PKCインヒビターと70%の一次構造上の類似性のあるトリプターゼTL2のシグナル伝達における役割を解析した。神経突起形成を誘導するプロテアーゼインヒビターの標的と思われるプロテアーゼを単離したところ、プロテアゾームの新しい活性であった。 2)細胞周期の制御におけるタンパク質分解の役割とその機構の解析:M期の終了をもたらすサイクリンのユビキチン依存的な分解の開始機構の解析がなされた。ツメガエル卵の受精におけるMosの分解に、MosN末端から2番目のプロリンおよびそれに隣接する3番目のセリンの脱リン酸化が必須であること、また、その分解にユビキチン-プロテゾーム系が関与していることが示された。 3)細胞外マトリックスの破壊・血管新生におけるプロテアーゼ系の役割の解析:MPP-9の大量産生は高転移能と関連があることおよび腫瘍由来のマクロファージは種々のメタロプロテアーゼの産生能を著明に亢進することを明確にした。基底膜の主要成分ラミニンがIV型コラーゲン分泌促進能と同時に増殖促進作用をもつこと、さらにその活性部位を同定した。
