LAK細胞の癌組織集積性を増強するサイトカインの精製とその遺伝子の同定

研究概要	癌化学療法後のマウス癌組織に一過性に産生され、養子移入されたリンホカイン活性化キラー(LAK)細胞を集積させるLAK-attractantの産生条件、産生機構を検討し、その精製と産生遺伝子の同定することを目的とした。まず、LAK-attractantの産生条件の検討から、これが非治療の癌組織には検出限界以下であるが、サイクロフォスファマイド、アドレアマイシンなど多種の抗癌剤による化学療法後5日目をピークとして一過性に産生されることを確認した。このLAK-attractantの産生条件は移入LAK細胞の癌組織集積性の増強条件と一致した。LAK-attractant活性は56℃30分熱処理には安定であるが、95℃5分処理、pH2の酸処理で消失した。これら諸性状により、LAK-attractantが一種のサイトカインであると推察された。一方、既知サイトカインIL-1α、IL-6、IL-8、IFN-γ、TNF-αなどの産生遺伝子が癌組織で発現されていることがRT-PCR法で明らかになったが、化学療法によってその発現が上昇するものはなかった。さらに、既知サイトカインのリコンビナント標品にはLAK-attractant活性が認められなかった。従って、化学療法後とくに腫瘍組織に産生されるようになるLAK-attractantが未知の新しいサイトカインである可能性があり、現在その活性の濃縮を指標に精製分離の過程にある。LAK-attractantの本態が明らかになって癌組織に高く発現させることができれば、LAK細胞を用いた養子免疫療法の効果の著しい増強の可能性が期待できる。
