転写調節機構の解析-調節因子と基本因子との相互作用を中心とした解析

研究概要	転写調節因子による転写活性化機構を理解するためには、転写調節因子と転写基本因子間の分子間相互作用の解析およびその結果に基づいた綿密な考察が必要である。本研究においては、1988年に私自身がモデルを提唱して以来、これまでに明らかとなってきた転写調節機構論の概念に沿って、TATAボックス結合因子TFIIDおよび転写基本因子TFIIBに関する機能および構造の解析を特に行ない、以下に示す結果を得ることができた。これらの結果は、転写調節機構を考える上で極めて重要な知見を含むものと確信している。 1.TFIIDサブユニットの単離 2.TFIIDサブユニットのひとつが細胞周期G1→S期に関与するCCG1であることの発見 3.TFIIDサブユニットp110のcDNA単離とその性状解析 4.TFIIDサブユニットp85のcDNA単離および、ある種のG蛋白内のWD40リピート構造を持つことの発見 5.TBPのRNAポリメラーゼI,III系への関与 6.TBPのTATA1essプロモーターへの結合活性 7.転写基本因子TFIIBの構造と機能の関連解析 8.転写基本因子TFIIFサブユニットRAP74のcDNA単離
