マウス乳癌プロウイルスの自己スーパー抗原としての生物学的意義

研究概要

○新しいマウス乳癌プロウイルス(Mtv-48,50,51)を見いだし、Mtv-48がVβ2T細胞のdeletionと、Mtv-50がVβ6/8.1T細胞のdeletionとco-segregateすることを明らかにした。さらに、Mtv-48およびMtv-50の末端反復配列の塩基配列により、C末端にVβ特異性を決定する領域があることが推定された。
次に、個体発生における胸腺内での自己スーパー抗原反応性T細胞のclonal deletionの時期とMtv遺伝子発現の時期を比較したところ、Mtv-50の発現の遅れに伴って、Vβ6/8.1T細胞の胸腺内clonal deletionが遅れることが明らかとなった。
CS及びII-TES由来の母乳より、外来性マウス乳癌ウイルスを同定し、これらが、Vβ2及びVβ14T細胞を選択的に増殖させた後に細胞死に陥らせることを明らかにした。CD4 knock outマウスでは、これら外来性乳癌ウイルスの感染症は起こらず、CD4^+T細胞が感染症の進展に重要であることが明らかとなった。
○AIDSのマウスモデルであるMuLV感染症において、Vβ11T細胞が選択的にclonal anergyに陥っていることを見いだし、レトロウイルス感染症において、スーパー抗原が病態形成に重要な役割を担っている可能性が考えられた。
